SEP-228432 is a small molecule drug developed as a triple reuptake inhibitor, targeting the serotonin transporter (SERT), norepinephrine transporter (NET), and dopamine transporter (DAT). Its mechanism of action involves inhibiting the reuptake of these three monoamine neurotransmitters, thereby increasing their synaptic concentrations. The drug was originally developed by Sepracor and later by Sumitomo Pharma America (formerly Sunovion Pharmaceuticals/Dainippon Sumitomo Pharma America). It has been investigated primarily for major depressive disorder and neuropathic pain, with prior development also in attention-deficit hyperactivity disorder. Clinical development reached phase 1 trials in healthy volunteers but did not progress further for its main indications[1][2][5][7].
