sEphB4-HSA is a recombinant fusion protein composed of the extracellular domain of human Ephrin type-B receptor 4 (EphB4) fused to full-length human serum albumin (HSA). It acts as a soluble decoy receptor that binds specifically to the membrane-bound ligand ephrin-B2, thereby blocking its interaction with endogenous cell surface receptors such as EphB4 and EphA3. This blockade disrupts bidirectional signaling between tumor cells and tumor vasculature, leading to reduced angiogenesis, inhibition of tumor growth, and potentially enhanced immune cell infiltration into tumors. The drug is being developed primarily for various solid tumors and hematologic malignancies including acute myeloid leukemia, bladder cancer, head and neck cancer, prostate cancer, Kaposi's sarcoma, myelodysplastic syndromes, non-small cell lung cancer, urogenital cancers, chronic myelomonocytic leukemia, and other solid tumors. Key developers include VasGene Therapeutics in collaboration with the National Cancer Institute (USA) and University of Southern California.
