sEphB4-HSA + chemotherapy is a combination therapy consisting of the recombinant fusion protein sEphB4-HSA and standard chemotherapeutic agents. sEphB4-HSA is an albumin-bound extracellular fragment of the EphB4 receptor, designed to inhibit the interaction between EphB4 and its ligand, EphrinB2. This blockade disrupts bidirectional signaling that promotes tumor angiogenesis, growth, and immune evasion. By inhibiting this pathway, sEphB4-HSA suppresses downstream oncogenic signals (such as KRAS and PI3K) and enhances immune cell infiltration into tumors. When combined with standard chemotherapies like cisplatin or carboplatin (for various cancers including pancreatic cancer [PC] and bladder cancer [BC]), this regimen aims to improve disease control by targeting both tumor cell proliferation (via chemotherapy) and tumor microenvironment/angiogenesis (via sEphB4-HSA)[1][4]. The combination has been studied in phase Ib/II trials for advanced solid tumors.
