This is an investigational CAR-T cell therapy regimen developed by Beijing Boren Hospital for the treatment of relapsed or refractory B-cell acute lymphoblastic leukemia (B-ALL), particularly in pediatric patients. The therapy consists of the sequential administration of two distinct chimeric antigen receptor (CAR) T-cell products: an anti-CD19 CAR-T followed by an anti-CD22 CAR-T, typically at an interval of 1.5 to 6.5 months. Both products utilize second-generation CAR constructs featuring 4-1BB costimulatory domains and CD3ζ signaling domains, delivered via lentiviral vectors into either patient-derived or donor-derived T-cells. This dual-targeting sequential approach is designed to improve long-term relapse-free survival and mitigate the risk of antigen-negative relapse, a common limitation of single-target CAR-T therapies.
