Overview

Seribantumab is a fully human monoclonal antibody (IgG2) developed for the treatment of cancer, particularly solid tumors harboring NRG1 gene fusions. It targets the human epidermal growth factor receptor 3 (HER3/ErbB3), binding to its extracellular domain and blocking neuregulin 1 (NRG1) from activating the receptor. This inhibition prevents downstream signaling through pathways such as PI3K-AKT and MAPK, which are involved in tumor cell survival and proliferation. Seribantumab was originally developed by Merrimack Pharmaceuticals and Sanofi, with later development by Elevation Oncology. The drug has been investigated in multiple clinical trials for various cancers including non-small cell lung cancer, ovarian cancer, breast cancer, colorectal cancer, and other solid tumors.
