Overview

**Seribantumab + paclitaxel** is a combination therapy under investigation primarily for **platinum-resistant or -refractory ovarian cancer**. Seribantumab is a fully human immunoglobulin G2 monoclonal antibody that targets human epidermal growth factor receptor 3 (HER3/ErbB3), blocking the interaction with its ligand heregulin (HRG/NRG1), and thereby inhibiting downstream HRG-mediated ErbB3 signaling and inducing receptor downregulation. Paclitaxel is a standard chemotherapeutic agent that promotes microtubule stabilization, inhibiting cell division. The combination was studied in a phase II randomized clinical trial versus paclitaxel alone. While the overall unselected patient population showed no improvement in progression-free survival or overall survival, biomarker analyses suggested that patients with tumors expressing high HRG and low HER2 benefited from the addition of seribantumab. The drug combination has not received regulatory approval for any indication[1][3][6][7].
