Series 1 Compounds (also known as the KLS-1 Series) are a class of ruthenium-based small molecule antibiotics being developed by MetalloBio. These compounds, which include the lead candidate KLS-1001, represent a novel class of metallo-antimicrobials designed to combat multi-drug resistant (MDR) pathogens. They exhibit a unique bactericidal mechanism of action that involves the disruption of bacterial cell membranes and potential interaction with bacterial DNA, providing broad-spectrum activity against both Gram-negative and Gram-positive ESKAPE pathogens (including *Staphylococcus aureus*, *Klebsiella pneumoniae*, and *Pseudomonas aeruginosa*). A distinguishing feature of the Series 1 program is its ability to penetrate and disrupt bacterial biofilms, which are significant contributors to chronic and hospital-acquired infections. The program is currently in preclinical development for indications including complicated urinary tract infections (cUTIs), hospital-acquired pneumonia (HAP), and ventilator-associated pneumonia (VAP).
