Serlopitant is a small molecule drug that acts as a highly selective antagonist of the neurokinin 1 (NK1) receptor, also known as the substance P receptor. It was originally developed by Merck and later licensed to Menlo Therapeutics for further development. Serlopitant has been investigated primarily for the treatment of pruritus (itch), particularly in conditions such as prurigo nodularis and chronic pruritus of unknown origin. The mechanism of action involves blocking the NK1 receptor, thereby inhibiting substance P-mediated signaling pathways implicated in itch transmission. Clinical trials have shown that serlopitant can significantly reduce itch intensity in patients with chronic refractory pruritic conditions and is generally well tolerated[2][4][5][6][7][8].
