Overview

SERPINE1 short hairpin RNA is an experimental RNA interference (RNAi) therapeutic designed to silence the expression of the *SERPINE1* gene, which encodes Plasminogen Activator Inhibitor-1 (PAI-1). PAI-1 is a key regulator of the fibrinolytic system and is frequently overexpressed in various malignancies, where it contributes to tumor progression, cell proliferation, and metastasis. The shRNA molecule functions by binding to complementary sequences on the SERPINE1 mRNA, leading to its degradation and a subsequent reduction in PAI-1 protein levels. Research indicates that the effects of SERPINE1 knockdown are context-dependent; while it has been shown to inhibit growth and invasion in breast cancer and glioma models by modulating the ERK/p38 signaling axis, it may paradoxically promote proliferation in certain melanoma contexts through the modulation of p53/SMAD3 regulators.
