SERPINE2 short hairpin RNA (shRNA) is an RNA interference-based therapeutic candidate designed to specifically silence the expression of the SERPINE2 gene. SERPINE2, a member of the serpin superfamily, is significantly upregulated in melanoma tissues and is associated with cell growth, epithelial-mesenchymal transition (EMT), and immune evasion. By targeting SERPINE2 mRNA for degradation, this shRNA inhibits cell viability, proliferation, and invasion. Mechanistically, silencing SERPINE2 has been shown to modulate the DDX3X/MITF/p21 signaling pathway, inducing cell cycle arrest in the G0/G1 phase and promoting apoptosis. Furthermore, downregulation of SERPINE2 enhances CD8+ T cell infiltration and activation, suggesting its potential as a biomarker and therapeutic target for melanoma immunotherapy.
