Serplulimab + etoposide + carboplatin is a combination immunotherapy and chemotherapy regimen being investigated for the treatment of small-cell lung cancer (SCLC) that has transformed from EGFR-mutated non-small cell lung cancer (NSCLC). Serplulimab is a humanized monoclonal antibody that functions as an immune checkpoint inhibitor by targeting the programmed cell death protein 1 (PD-1) receptor, thereby blocking its interaction with ligands PD-L1 and PD-L2 to restore T-cell anti-tumor activity. Etoposide is a small molecule topoisomerase II inhibitor that induces DNA strand breaks, while carboplatin is a platinum-based alkylating agent that forms DNA cross-links, both providing cytotoxic effects. This specific combination is the subject of an investigator-initiated Phase II clinical trial (NCT05957510) sponsored by the Guangdong Association of Clinical Trials to evaluate its safety and efficacy in patients with transformed SCLC after prior EGFR-targeted therapy.
