Serplulimab + FOLFOXIRI is an investigational combination therapy for cancer, particularly metastatic colorectal cancer. Serplulimab is a humanized monoclonal antibody targeting programmed cell death protein 1 (PD-1), functioning as an immune checkpoint inhibitor to enhance T-cell mediated anti-tumor responses. It is developed by Henlius and marketed under the brand name HANSIZHUANG. FOLFOXIRI is a chemotherapy regimen consisting of four drugs: folinic acid (leucovorin), fluorouracil, oxaliplatin, and irinotecan. This regimen acts through multiple mechanisms—antimetabolite activity from fluorouracil, DNA crosslinking from oxaliplatin, topoisomerase I inhibition from irinotecan, and modulation of fluorouracil toxicity by leucovorin—to inhibit tumor growth and proliferation[6][5]. The combination aims to leverage both immunotherapy and cytotoxic chemotherapy for enhanced efficacy in advanced or metastatic colorectal cancer.
