This combination regimen is a multi-modal therapeutic approach being evaluated for the first-line treatment of metastatic or locally advanced pancreatic ductal adenocarcinoma (PDAC). It consists of the anti-PD-1 monoclonal antibody serplulimab, the chemotherapy agents gemcitabine and nab-paclitaxel (collectively known as the GnP regimen), and stereotactic body radiation therapy (SBRT). Serplulimab, developed by Henlius, functions as a checkpoint inhibitor to enhance the immune system's anti-tumor response by blocking the PD-1 pathway. Gemcitabine and nab-paclitaxel provide a cytotoxic backbone that inhibits DNA synthesis and microtubule function, respectively. SBRT is employed as a high-dose local treatment intended to induce immunogenic cell death and potentially synergize with the immunotherapy component. The regimen is currently being investigated in Phase II and Phase III clinical trials sponsored by West China Hospital of Sichuan University.
