Overview

SET-M33 is a synthetic, tetra-branched antimicrobial peptide (AMP) being developed by SetLance for the treatment of severe bacterial infections, including sepsis, pneumonia, and skin infections caused by multidrug-resistant (MDR) Gram-negative and Gram-positive pathogens. Synthesized as a D-stereoisomer to enhance resistance against protease and peptidase degradation in biological fluids, SET-M33 operates via a two-step mechanism of action: it binds with high affinity to bacterial lipopolysaccharide (LPS) and subsequently disrupts the bacterial outer membrane, leading to rapid cell lysis. In addition to its direct bactericidal activity, SET-M33 exhibits potent anti-inflammatory properties by neutralizing LPS and preventing the release of pro-inflammatory cytokines. The drug is currently in preclinical development, with completed toxicological and efficacy evaluations in animal models.
