SETD8 shRNA is a short hairpin RNA (shRNA) designed to silence the expression of the SETD8 gene (also known as KMT5A or PR-Set7). SETD8 is the sole methyltransferase responsible for the mono-methylation of histone H4 at lysine 20 (H4K20me1), an epigenetic mark associated with transcriptional activation and cell cycle regulation. In the context of Acute Myeloid Leukemia (AML), SETD8 knockdown via shRNA has been shown to inhibit cell proliferation and induce apoptosis by downregulating the expression of CXCR4, a critical receptor for leukemia cell survival and infiltration. This genetic intervention serves as a tool for validating SETD8 as a therapeutic target in hematological malignancies.
