Overview

Setmelanotide is a synthetic peptide drug that acts as an agonist of the melanocortin 4 receptor (MC4R). It is indicated for chronic weight management in patients with obesity due to rare genetic conditions affecting the MC4R pathway, specifically pro-opiomelanocortin (POMC) deficiency, proprotein convertase subtilisin/kexin type 1 (PCSK1) deficiency, leptin receptor (LEPR) deficiency, and Bardet-Biedl syndrome (BBS). Setmelanotide works by activating MC4R in the hypothalamus to reduce appetite and increase energy expenditure. Unlike some other MC4R agonists, it does not significantly increase heart rate or blood pressure. The drug was developed by Ipsen and Rhythm Pharmaceuticals and is administered as a once-daily subcutaneous injection[2][3][5][6].
