Overview

This is a combination of three agents: sevuparin sodium, atovaquone, and proguanil. Atovaquone and proguanil are antimalarial drugs used together to treat and prevent malaria caused by *Plasmodium falciparum*. Atovaquone inhibits the mitochondrial electron transport chain in the parasite, while proguanil acts as a biguanide that enhances the effect of atovaquone on mitochondrial membrane potential; its metabolite cycloguanil also inhibits dihydrofolate reductase but this is not central to their synergy[1][6]. Sevuparin sodium is an experimental drug being studied as an adjunctive therapy for severe malaria; it is a modified heparinoid designed to block sequestration of infected red blood cells in microvasculature, potentially improving outcomes when combined with standard antimalarial therapies[4]. The triple combination has been investigated for improved efficacy against severe or complicated malaria.
