Overview

SF2a-TT15 is a synthetic carbohydrate-based conjugate vaccine candidate developed to protect against *Shigella flexneri* serotype 2a (SF2a), a major cause of shigellosis and diarrheal disease, particularly in low-income countries. The vaccine consists of a synthetic 15mer oligosaccharide that mimics the O-antigen of *S. flexneri* 2a, covalently linked to tetanus toxoid (TT) as the carrier protein. This design aims to induce robust immune responses by presenting both B-cell and T-cell epitopes. Clinical studies have shown that SF2a-TT15 is safe, well tolerated, and highly immunogenic in healthy adults, inducing strong IgG and IgA antibody responses as well as long-lived memory B cells specific for SF2a LPS. The immune response has been observed to persist for at least three years post-vaccination. The vaccine is being developed by Institut Pasteur.
