Overview

Sf2aWC is a formalin-inactivated, whole-cell oral vaccine candidate designed to prevent shigellosis caused by *Shigella flexneri* 2a. Originally developed by the Walter Reed Army Institute for Research (WRAIR) and currently being advanced by PATH, the vaccine targets the lipopolysaccharide (LPS) antigen on the bacterial surface. It is designed to induce mucosal and systemic immunity, including anti-LPS antibodies in serum and feces. Clinical evaluation includes testing the vaccine alone or in combination with the double mutant *E. coli* heat-labile toxin (dmLT) adjuvant to enhance immunogenicity and protective efficacy against gastrointestinal infection.
