SFELNVs (Sophora flavescens-derived exosome-like nanovesicles) are plant-derived nanotherapeutic agents extracted from the roots of the medicinal plant *Sophora flavescens*. These vesicles are designed for oral administration and demonstrate inherent targeting to macrophages within the inflamed gastrointestinal tract. SFELNVs exert anti-inflammatory effects by promoting M2 macrophage polarization, a process mediated by the delivery of miR4371c, and by suppressing the expression of pro-inflammatory cytokines including TNF-α, IL-1β, and IL-6. Beyond their intrinsic therapeutic activity, SFELNVs serve as a biocompatible delivery platform for small molecules like CX5461, enhancing drug concentration at the site of inflammation. Preclinical studies in DSS-induced colitis models indicate that SFELNVs can significantly ameliorate disease severity and promote mucosal healing with high safety and stability.
