Overview

sFlt1 adenovirus (also known as Ad-sFlt-1 or AdsFlt) is a recombinant adenoviral gene therapy vector designed to express soluble fms-like tyrosine kinase-1 (sFlt-1), which is the soluble extracellular domain of vascular endothelial growth factor receptor 1 (VEGFR1). The expressed sFlt-1 protein acts as a secreted decoy receptor that binds with high affinity to vascular endothelial growth factor A (VEGF-A) and placental growth factor (PlGF). By sequestering these pro-angiogenic ligands, sFlt-1 prevents their interaction with cell-surface receptors such as VEGFR1 and VEGFR2, thereby inhibiting VEGF-driven angiogenesis and reducing vascular permeability. In preclinical research, sFlt1 adenovirus is widely utilized to induce a preeclampsia-like state in animal models to study the resulting cardiovascular and renal complications. It has also been investigated as a potential anti-angiogenic therapeutic strategy for various cancers, including ovarian, breast, and colorectal carcinomas, as well as inflammatory conditions like rheumatoid arthritis. It is currently used strictly as a research tool and has not advanced to clinical development under a commercial sponsor.
