SFV-enhanced interleukin-12 is a gene therapy construct in which interleukin-12 (IL-12), a potent pro-inflammatory cytokine with antitumor and antiviral properties, is delivered via an engineered Semliki Forest Virus (SFV) vector. The SFV vector serves as a delivery system to express high levels of IL-12 in target tissues, leveraging viral infection to induce both innate and adaptive immune responses. The antitumor activity is primarily mediated through activation and expansion of cytotoxic CD8+ T cells and natural killer (NK) cells, increased production of interferon-gamma (IFN-γ), and reshaping of the tumor microenvironment toward a more immunogenic phenotype. This approach has demonstrated potent antitumoral efficacy and antiviral responses in preclinical models. The SFV vector itself is a self-replicating RNA virus vector designed for short-term, high-level transgene expression. Clinical development status appears to be preclinical or early research[4].
