Overview

SG-242 is an **autologous CAR-T cell therapy** under preclinical development by Sana Biotechnology. The therapy is engineered to target both **CD19** (a B-cell surface antigen) and **CD4** (a T-cell surface antigen), employing a chimeric antigen receptor that mediates cell-specific immunologic cytotoxicity. Its primary indications are **chronic lymphocytic leukemia** and **non-Hodgkin lymphoma**, with further interest in acute lymphoblastic leukemia. The core mechanism involves both **CD19 inhibition** (via B-lymphocyte antigen CD19) and **CD4 modulation** (via T-cell surface antigen CD4), enabling targeted elimination of malignant or dysfunctional B and T lymphocytes. SG-242 is a cellular immunotherapy product, relying on patient-derived (autologous) T lymphocytes genetically modified to express the chimeric receptor, and is not an in vivo gene therapy. All current development is preclinical and there is no approval or late-stage clinical trial for this therapy as of the most recent data[1].
