Overview

SG-5-00455 is an investigational **engineered bacterial cellular therapy** being developed by Second Genome for inflammatory bowel disease, including ulcerative colitis and Crohn's disease. The product uses an engineered *Lactococcus lactis* live biotherapeutic delivery system to provide oral, non-systemic delivery of the microbiome-derived protein SG-2-0776 directly in the gut. SG-2-0776 is designed to bind and inhibit plasminogen activator inhibitor 1 and plasminogen activator inhibitor 2, with the intended effect of restoring epithelial barrier function, promoting mucosal healing, improving dysregulated tissue repair, and reducing fibrosis associated with intestinal inflammation. Preclinical colitis data described reductions in inflammatory pathology and improvement in fibrosis- and repair-associated gene expression, and the program was reported to be in IND-enabling development.
