Overview

SG1827 is a bispecific monoclonal antibody (BsAb) developed by Hangzhou Sumgen Biotech, designed to treat advanced solid tumors expressing Claudin 18.2 (CLDN18.2). The molecule acts as a T-cell engager, featuring one arm that binds to CLDN18.2—a tight junction protein frequently overexpressed in gastric, gastroesophageal junction, and pancreatic cancers—and another arm that binds to the CD3 epsilon subunit of the T-cell receptor complex. By bridging these two targets, SG1827 promotes the formation of an immunological synapse, leading to the activation and proliferation of cytotoxic T-cells and the subsequent destruction of CLDN18.2-positive tumor cells. SG1827 is currently being evaluated in a Phase I clinical trial (NCT05948605) to determine its safety, tolerability, and preliminary efficacy in patients with refractory or resistant advanced solid tumors.
