SG600-p53 is a triple-regulated conditionally replicating adenovirus (CRAd) developed by Vector Gene Technology (also known as Shenzhen SiBiono Gene Tech) for the treatment of solid tumors. It functions as a dual-action therapeutic, combining oncolytic virotherapy with gene therapy. The virus is engineered for tumor-selective replication through two distinct regulatory mechanisms: the E1a gene (containing a 24-nucleotide deletion in the CR2 region) is controlled by the human telomerase reverse transcriptase (hTERT) promoter, and E1b gene expression is regulated by the hypoxia response element (HRE). Additionally, the vector carries a p53 gene expression cassette under the control of a cytomegalovirus (CMV) promoter. This design ensures that the virus selectively replicates in and lyses telomerase-positive and hypoxic tumor cells while simultaneously overexpressing the p53 tumor suppressor protein to induce apoptosis and cell-cycle arrest in cancer cells.
