Overview

SGD-1882 is a cytotoxic pyrrolobenzodiazepine (PBD) dimer that functions as a DNA minor-groove crosslinking agent and serves as a payload (cytotoxic warhead) in antibody-drug conjugates (ADCs)[1][2][3][5][11][14]. By crosslinking DNA in target cells, SGD-1882 effectively disrupts DNA replication and transcription, leading to cell cycle arrest in the G2/M phase and rapid induction of apoptosis[2][4][14][15]. As a non-MDR1 substrate and a highly potent DNA alkylator, it is currently used in preclinical and clinical evaluation with anti-CD33, anti-CD19, and anti-CD70 ADCs for targeting hematological malignancies, particularly acute myeloid leukemia (AML)[1][2][4][15]. SGD-1882 is not used as a standalone therapeutic, but only as a payload in ADCs.
