SGI-1215 is a small molecule inhibitor of Janus kinase 2 (JAK2), a non-receptor tyrosine kinase that plays a critical role in the signaling pathways of various cytokines and growth factors. Developed by SuperGen (now Astex Pharmaceuticals), SGI-1215 was designed to target myeloproliferative neoplasms (MPNs), such as polycythemia vera, essential thrombocythemia, and primary myelofibrosis. These disorders are frequently characterized by the JAK2 V617F mutation, which causes constitutive activation of the JAK-STAT pathway, leading to the overproduction of blood cells. SGI-1215 acts as an ATP-competitive inhibitor, binding to the kinase domain of JAK2 to prevent phosphorylation and downstream signaling, thereby inhibiting the proliferation of malignant hematopoietic cells and inducing apoptosis. While SGI-1215 demonstrated potent activity in preclinical models, its clinical development was eventually discontinued in favor of other candidates or due to the competitive landscape of JAK inhibitors.
