Overview

SGI-498 is a potent, small molecule inhibitor of Polo-like kinase 1 (PLK1), a critical serine/threonine kinase that regulates multiple stages of the mitotic phase of the cell cycle. Developed by SuperGen (now part of Astex Pharmaceuticals), SGI-498 was designed as an antineoplastic agent to target the ATP-binding site of PLK1, thereby disrupting centrosome maturation, spindle assembly, and chromosome segregation. Inhibition of PLK1 by SGI-498 leads to mitotic arrest at the G2/M phase and subsequently induces apoptosis in rapidly proliferating cancer cells. PLK1 is frequently overexpressed in various human malignancies, including solid tumors and hematological cancers, and its expression levels often correlate with poor clinical prognosis, making it a high-value target for precision oncology. Preclinical evaluations demonstrated that SGI-498 possesses favorable oral bioavailability and significant anti-proliferative activity across multiple tumor cell lines.
