Overview

SGI-AXL-277 is a small molecule inhibitor of the AXL receptor tyrosine kinase, featuring a pyrrolopyrimidine scaffold. It was developed by SuperGen (which later merged with Astex Pharmaceuticals). AXL is a member of the TAM (Tyro3, Axl, Mer) receptor tyrosine kinase family and is frequently overexpressed in various malignancies, where it promotes tumor cell survival, epithelial-to-mesenchymal transition (EMT), and resistance to chemotherapy and targeted agents. SGI-AXL-277 was reported to be in the lead optimization and biological testing phases of preclinical development around 2011, specifically targeting solid tumors. However, there has been limited information regarding its clinical progression or current status in the pipeline since that time.
