Overview

SGK1-I is a novel, potent small molecule inhibitor of Serum and Glucocorticoid-Regulated Kinase 1 (SGK1) being developed by Thryv Therapeutics. SGK1 is a PI3-kinase-dependent kinase that plays a critical role in regulating renal sodium homeostasis, inflammation, and cardiac fibrosis. The compound is designed to address the pathophysiology of heart failure with preserved ejection fraction (HFpEF) and atrial fibrillation by mitigating hypertension-induced pressure overload and adverse cardiac remodeling. Preclinical studies in Dahl salt-sensitive rat models have demonstrated that oral administration of SGK1-I dose-dependently reduces systolic and diastolic arterial blood pressure and improves left ventricular morphology, including posterior wall thickness and mass-to-tibia length ratios.
