SGN-101 is an investigational 0.05% micellar nanoparticle (MNP) ophthalmic emulsion of cyclosporine A, developed by SGN Nanopharma for the treatment of chronic dry eye disease (keratoconjunctivitis sicca). Utilizing a proprietary nanotechnology platform, SGN-101 delivers cyclosporine in neutral to positively-charged nanoparticles (150-170 nm) designed to enhance penetration into deep ocular tissues and reduce surface irritation compared to traditional oil-based formulations like Restasis. In 2024, SGN Nanopharma entered a collaboration with Eyenovia to develop SGN-101 as a drug-device combination product using the Optejet microdosing dispenser, which employs Microdose Array Print (MAP) technology for precise, low-velocity delivery. Clinical data from a randomized controlled trial of 155 subjects demonstrated that SGN-101 was statistically superior to Restasis (cyclosporine 0.05%) in improving dry eye symptoms starting at week 4 of treatment.
