SGN-15 is an antibody-drug conjugate (ADC) composed of the chimeric monoclonal antibody BR96 chemically linked to the chemotherapeutic agent doxorubicin via an acid-labile hydrazone linker. The antibody component targets the Lewis Y (Le(y)) antigen, a carbohydrate antigen highly expressed on many solid tumors including non-small cell lung cancer (NSCLC), breast cancer, colorectal cancer, ovarian cancer, and prostate cancer. Upon binding to tumor cells expressing Le(y), SGN-15 is internalized and releases doxorubicin in the acidic environment of tumor cells. Doxorubicin then exerts cytotoxic effects by generating reactive oxygen species, inhibiting RNA synthesis, and inhibiting type II DNA topoisomerase activity. SGN-15 was developed as a targeted therapy to deliver chemotherapy directly to tumor cells while minimizing systemic toxicity.
