SGN-70 is a humanized IgG1 monoclonal antibody that targets CD70 (also known as CD27L), a member of the tumor necrosis factor (TNF) superfamily. CD70 is aberrantly expressed on various hematologic malignancies, such as Hodgkin lymphoma, non-Hodgkin lymphoma, and multiple myeloma, as well as certain carcinomas like renal cell carcinoma, while its expression on normal cells is highly restricted. Developed by Seattle Genetics (now Seagen), SGN-70 exerts its antitumor effects through Fc-dependent effector functions, including antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and antibody-dependent cellular phagocytosis (ADCP). Although SGN-70 demonstrated efficacy in preclinical xenograft models, its clinical development was largely superseded by antibody-drug conjugate (ADC) derivatives targeting the same antigen, such as SGN-75.
