SGN-CD19B is an investigational antibody-drug conjugate (ADC) designed for the treatment of B-cell malignancies, including relapsed or refractory non-Hodgkin lymphoma (NHL) and B-cell acute lymphoblastic leukemia. It consists of an anti-CD19 monoclonal antibody linked to a pyrrolobenzodiazepine (PBD) dimer, a highly potent DNA cross-linking cytotoxic agent. The ADC specifically targets CD19, a cell surface antigen broadly expressed on normal and malignant B-cells. Upon binding to CD19-expressing cells, SGN-CD19B is internalized and trafficked to lysosomes where the PBD payload is released, inducing DNA damage that leads to G2/M cell cycle arrest and apoptosis in tumor cells. Preclinical studies have shown broad antitumor activity against various CD19-positive B-cell lines and durable tumor regressions in animal models. Clinical development has focused on relapsed/refractory aggressive B-cell NHL subtypes such as diffuse large B-cell lymphoma (DLBCL) and grade 3 follicular lymphoma[1][2][3][5][6].
