SGN-CD47M is an antibody-drug conjugate (ADC) developed for the treatment of solid tumors. It consists of a humanized monoclonal IgG1 antibody targeting the cell surface antigen CD47, which is overexpressed on many tumor cells and acts as a "don't eat me" signal to evade immune surveillance. The antibody is linked to novel masking peptides via protease-cleavable linkers that are selectively activated in the tumor microenvironment, aiming to reduce systemic toxicity and enhance antitumor activity. This design allows for preferential activation within tumors, potentially improving tolerability compared to unmasked anti-CD47 antibodies. The drug was initially developed by Seagen and entered phase 1 clinical trials for various solid tumors but has since been discontinued[2][5][6].
