SGN-CD48A is an investigational antibody-drug conjugate (ADC) developed for the treatment of multiple myeloma. It consists of a humanized monoclonal antibody targeting the cell surface antigen CD48, which is highly expressed on multiple myeloma cells. The antibody is conjugated to eight molecules of monomethyl auristatin E (MMAE), a potent microtubule-disrupting cytotoxic agent, via a proprietary PEGylated glucuronide linker designed to improve stability and drug delivery. Upon binding to CD48 on myeloma cells, SGN-CD48A internalizes and releases MMAE intracellularly, leading to G2/M cell cycle arrest and caspase-dependent apoptotic cell death. Preclinical studies demonstrated potent antitumor activity with minimal effects on normal lymphocytes. Clinical development included phase 1 trials in relapsed or refractory multiple myeloma[1][2][3][4][5][6].
