Overview

SGN1 is an investigational, first-in-class oncolytic bacterial therapy being developed by Guangzhou Sinogen Pharmaceutical Co., Ltd. [11, 16] It consists of a genetically modified, attenuated strain of *Salmonella typhimurium* (a modified version of the VNP20009 strain) engineered to overexpress the enzyme L-methioninase. [2, 4, 5] The therapy is designed based on the metabolic vulnerability of many cancers, which are dependent on an external supply of the amino acid L-methionine for growth. [3] When administered, SGN1 preferentially targets and accumulates in solid tumors. [2, 5] Within the tumor microenvironment, it releases L-methioninase, which breaks down L-methionine, thereby starving the cancer cells of this essential nutrient. [2, 3, 5] This targeted methionine deprivation leads to cell-cycle arrest and apoptosis of tumor cells, inhibiting tumor growth and metastasis. [2, 5] SGN1 is being evaluated in clinical trials for a broad range of advanced solid tumors, administered either by intravenous infusion or intratumoral injection. [3, 12]
