SGR-0320 is a preclinical-stage small molecule inhibitor of Leucine-rich repeat kinase 2 (LRRK2) being developed by Schrodinger for the treatment of neurological disorders. LRRK2 is a large multidomain protein with kinase and GTPase activities; mutations that increase its kinase activity are among the most common genetic risk factors for both familial and sporadic Parkinson's disease. SGR-0320 was identified and optimized using Schrodinger's physics-based computational platform to achieve high potency and selectivity while ensuring the blood-brain barrier penetration necessary for central nervous system activity. The program aims to modulate neurodegenerative pathways implicated in Parkinson's disease and potentially other neurological conditions.
