Overview

SGR-1505 is an investigational, oral, small molecule allosteric inhibitor of mucosa-associated lymphoid tissue lymphoma translocation protein 1 (MALT1). MALT1 is a key mediator of the NF-κB signaling pathway and plays a central role in the survival and proliferation of certain B-cell malignancies. By inhibiting MALT1 enzymatic activity, SGR-1505 demonstrates anti-proliferative effects in activated B cell-like diffuse large B cell lymphoma (ABC-DLBCL) cell lines, including those resistant to Bruton’s tyrosine kinase inhibitors. Preclinical studies show tumorostatic and regressive antitumor activity both as monotherapy and in combination with other agents such as ibrutinib or venetoclax. SGR-1505 is currently being evaluated in Phase 1 clinical trials for relapsed/refractory mature B-cell malignancies, including various non-Hodgkin lymphomas and chronic lymphocytic leukemia[1][2][3][4][5].
