SGR-4174 is a novel, potent, and selective small molecule inhibitor of SOS1 (Son of sevenless homolog 1), developed by Schrödinger for the treatment of cancers driven by KRAS mutations. SOS1 is a key activator in the RAS signaling pathway, which regulates cell proliferation and survival; its inhibition disrupts KRAS activation and downstream oncogenic signaling. Preclinical data demonstrate that SGR-4174 robustly suppresses RAS pathway activity and induces tumor cell death across multiple cancer types harboring diverse KRAS mutations as well as EGFR and SOS1 mutations. The compound shows dose-dependent target engagement, tumor growth inhibition as monotherapy, and enhanced tumor shrinkage when combined with MEK or KRAS inhibitors in models of pancreatic and non-small cell lung cancer. It is currently in preclinical development[1][2][3][7].
