Overview

SGR-4281 is a small-molecule inhibitor of the paracaspase MALT1 (Mucosa-associated lymphoid tissue lymphoma translocation protein 1), currently in preclinical development by Schrodinger for the treatment of inflammatory and immunological disorders. MALT1 is a key component of the CARMA1-BCL10-MALT1 (CBM) complex, which plays a critical role in transmitting signals from antigen receptors on T and B cells to the NF-κB pathway, a central regulator of immune and inflammatory responses. While Schrodinger's other MALT1 inhibitor, SGR-1505, is primarily positioned for oncology (specifically B-cell lymphomas), SGR-4281 is optimized with a profile intended for chronic dosing in immunology and inflammation. By inhibiting MALT1, SGR-4281 aims to modulate overactive immune pathways and reduce the production of pro-inflammatory cytokines.
