Overview

SGS-742 is an orally available small molecule antagonist of the gamma aminobutyric acid-B (GABA-B) receptor. It was developed as a potential treatment for cognitive disorders such as mild cognitive impairment, Alzheimer's disease, attention-deficit hyperactivity disorder (ADHD), epilepsy, and succinic semialdehyde dehydrogenase deficiency (SSADH-D). The drug acts by blocking GABA-B receptors in the central nervous system. Preclinical studies suggested that SGS-742 could improve attention, reaction time, visual information processing, and working memory in animal models. In clinical trials—including phase II studies—SGS-742 did not demonstrate significant efficacy for improving cognition or normalizing cortical excitability in patients with SSADH-D or other neurocognitive disorders. The compound is excreted unchanged in urine and has a half-life of approximately four hours[1][2][4][7].
