SGT-53 + temozolomide is a combination therapy consisting of SGT-53, a tumor-targeting nanomedicine carrying the p53 gene, and temozolomide, an alkylating agent. SGT-53 works by restoring p53 function in tumor cells, which can boost anti-tumor immunity and enhance treatment effectiveness. Temozolomide is a prodrug that undergoes spontaneous conversion at physiological pH to form its active metabolite, which then alkylates DNA, causing damage that leads to cancer cell death. This combination was investigated for the treatment of recurrent glioblastoma.
