SGW13 is an orally available, potent, and highly selective small molecule inhibitor of Aurora kinase A (AURKA). Aurora kinase A is a serine/threonine protein kinase that plays a pivotal role in the regulation of mitosis, specifically in centrosome maturation and the assembly of the mitotic spindle. Its overexpression is associated with centrosome amplification, chromosomal instability, and poor clinical outcomes in a variety of solid and hematological malignancies. By selectively inhibiting Aurora A over Aurora B, SGW13 is designed to induce mitotic arrest and trigger apoptosis in rapidly dividing cancer cells while potentially avoiding the neutropenia often associated with non-selective Aurora kinase inhibition. The drug is currently undergoing clinical investigation to evaluate its safety, tolerability, and pharmacokinetic profile in patients with advanced malignancies.
