SH-4-54 is a potent small molecule inhibitor of Signal Transducer and Activator of Transcription (STAT) proteins, specifically targeting STAT3 and STAT5. It functions by directly binding to the SH2 domain of STAT3 (KD = 300 nM) and STAT5 (KD = 464 nM), thereby preventing their phosphorylation, dimerization, and subsequent nuclear translocation. This inhibition disrupts the transcription of genes involved in cell survival, proliferation, and self-renewal. Developed by researchers at the University of Hawaii Cancer Center, SH-4-54 has demonstrated significant preclinical activity against various malignancies, most notably glioblastoma. Its ability to cross the blood-brain barrier makes it a particularly promising candidate for treating brain tumors and targeting brain tumor-initiating cells (BTSCs). Preclinical studies have also indicated potential efficacy in bladder, pancreatic, breast, and prostate cancers.
