Overview

SH2442 is an orally bioavailable, small molecule selective agonist of the thyroid hormone receptor beta (THR-β), developed by Nanjing Sanhome Pharmaceutical. It is primarily investigated for the treatment of non-alcoholic fatty liver disease (NAFLD) and non-alcoholic steatohepatitis (NASH). THR-β is the predominant thyroid hormone receptor isoform in the liver, responsible for regulating metabolic pathways including cholesterol lowering and triglyceride reduction. By selectively activating THR-β while avoiding the THR-α isoform (which is associated with heart rate and bone density effects), SH2442 aims to reduce hepatic steatosis and improve metabolic health. Clinical trials have progressed to Phase 2 in China, focusing on its efficacy in reducing liver fat and improving histological markers of NASH.
