Overview

SH3602 (Augrecerti) is an orally bioavailable, highly selective, and irreversible small molecule inhibitor of fibroblast growth factor receptor 4 (FGFR4), developed by Nanjing Sanhome Pharmaceutical. It is specifically designed to target the FGF19-FGFR4 signaling axis, which is a critical driver of tumor growth and survival in a subset of patients with hepatocellular carcinoma (HCC). By covalently binding to the FGFR4 kinase domain, SH3602 inhibits downstream signaling pathways such as MAPK/ERK and PI3K/AKT, thereby suppressing oncogenic activity and tumor cell proliferation. The drug is currently in Phase 1 clinical trials for the treatment of advanced solid tumors and FGF19-overexpressing liver cancer, where it aims to provide a targeted therapeutic option for patients with specific molecular alterations in the FGFR pathway.
