Overview

SH5-07 is a small molecule inhibitor of Signal Transducer and Activator of Transcription 3 (STAT3). It functions by targeting the SH2 domain of STAT3, which prevents its phosphorylation, dimerization, and subsequent nuclear translocation and DNA binding. In preclinical studies, particularly in 3D tumor models of bladder cancer, SH5-07 has demonstrated the ability to reduce tumor spheroid size, decrease cell viability, and induce apoptosis. It also promotes the production of reactive oxygen species (ROS) and activates the cGAS-STING pathway, which may enhance immune modulation within the tumor microenvironment. The compound is being explored as a potential orally bioavailable therapeutic agent for muscle-invasive bladder cancer and other malignancies characterized by aberrant STAT3 signaling.
