Shattuck Labs is developing a pipeline of bispecific antibodies targeting Death Receptor 3 (DR3) to inhibit the TL1A/DR3 signaling pathway, a key driver of inflammation and immune dysregulation. These preclinical candidates are designed to block the DR3/TL1A axis while simultaneously targeting a second, biologically relevant pathway for the treatment of autoimmune and inflammatory conditions, such as inflammatory bowel disease (IBD). By targeting the membrane-restricted DR3 receptor rather than the soluble TL1A ligand, these antibodies aim to avoid the formation of large immune complexes and the high immunogenicity observed with TL1A-directed therapies. As of early 2026, the lead bispecific program has entered IND-enabling activities, with further details on targets and preclinical data expected in the first half of 2026.
